Upstream Intensification – Enabling Perfusion Processes with Cell Retention Technologies

Perfusion processes offer key advantages to fed-batch processes and are now possible thanks to recent advances in cell retention technologies, cell line, and media development. What features should such technologies have to achieve performance, scalability, and cell health? In this study, new perfusion technologies – cell retention device and specially designed expansion media- are applied in a perfused N-1 process prior to a fed-batch production bioreactor. The effects of their use in the seed train are compared to a control process and evaluated for cell growth, titer, nutrients and metabolites, and product quality during the production process.
