Volume Reduction and Process Optimization with Cadence™ Inline Concentrator

Abstract

Traditional tangential flow filtration (TFF) is one of the most widely used technologies in downstream bioprocessing.  It can be used for concentration or diafiltration and has traditionally been operated in batch mode.  Operating in batch mode has some disadvantages in terms of overall process economics, for example large volumes of process fluid must be handled and recirculation takes time.   

An alternative to traditional batch mode is single-pass tangential flow filtration (SPTFF) which eliminates the TFF recirculation loop.  This minimizes mixing or foaming issues and exposure of the product to shear forces, making it ideal for processing of fragile and shear-sensitive molecules.   
 
For concentration applications, single-pass feed flow rates are lower which minimizes the hold-up volume and maximizes product recovery.  It also allows higher final concentrations to be achieved as minimum working volumes are optimized. 

This application note details how, by using SPTFF, batch volume was reduced and space limitation issues were resolved in a concentration step for a highly viscous feed stream that consisted of a suspension of nanoparticles used in therapeutic drug delivery applications.
